Se ha estudiado la interacción entre antígenos ABO y microorganismos, incluidos los presentes en la microbiota, sobre la posible acción de antígenos y anticuerpos ABO en la susceptibilidad a enfermedades infecciosas. El objetivo de esta investigación fue determinar el título mínimo de la bacteria Escherichia coli capaz de sufrir la acción bactericida in vitro de los anticuerpos humanos anti-ABO. La selección de las muestras de sangre utilizadas se realizó mediante la aplicación de un cuestionario, fenotipado sanguíneo (un voluntario de cada fenotipo ABO) y la titulación de anticuerpos ABO. Se preparó una suspensión bacteriana (inoculo) y se agregó al suero de los voluntarios, seguido de la inoculación en Mueller Hinton Agar, luego de 24 horas, los resultados se leyeron e interpretaron con análisis por duplicado. No hubo diferencia significativa en la Prueba Bactericida entre las pruebas 1 y 2 en los grupos sanguíneos A, B, AB, O y Control Positivo. Hubo una diferencia significativa en el suero humano puro cuando se analizó el Grupo A x Control Positivo; Grupo B x Control Positivo; Grupo AB x Control Positivo y Grupo O x Control Positivo. No hubo diferencia significativa en las otras diluciones. Se concluye que los anticuerpos anti-ABO tienen efecto bactericida cuando existe una alta concentración de bacterias en el ambiente.


The interaction between ABO antigens and microorganisms, including those present in the microbiota, has been studied about the possible action of antigens and ABO antibodies in susceptibility to infectious diseases. This research aimed to determine the minimum titer of the Escherichia coli bacteria capable of undergoing in vitro bactericidal action of human anti-ABO antibodies. The selection of blood samples was performed through a questionnaire, blood phenotyping (one volunteer of each ABO phenotype), and the titration of ABO antibodies. A bacterial suspension (inoculum) was prepared and added to the serum of the volunteers, followed by inoculation in Mueller Hinton Agar. After 24 hours, the results were read and interpreted with duplicate analysis. There was no significant difference in the bactericidal test between tests 1 and 2 in blood groups A, B, AB, O, and Positive Control. There was a significant difference in pure human serum when Group A x Positive Control was analyzed, Group B x Positive Control, Group AB x Positive Control, and Group O x Positive Control. There was no significant difference in the other dilutions. It is concluded that anti-ABO antibodies have a bactericidal effect when there is a high concentration of bacteria in the environment.

Harmful algal blooms (HABs), caused primarily by nutrient input from agricultural runoff, are a threat to freshwater systems worldwide, and are further predicted to increase in size, frequency, and intensity due to climate change. HABs occur annually in the Western Basin of Lake Erie (Ohio, USA), and these blooms become toxic when dominated by cyanobacteria that produce the liver toxin microcystin. Although we are making substantial inroads toward understanding how microcystin affects human health, less is known about effects of microcystin on wildlife exposed to HABs. Wetland-associated songbirds (barn swallows, Hirundo rustica, and red-winged blackbirds, Agelaius phoeniceus) and reptiles (Northern watersnakes, Nerodia sipedon, and painted turtles, Chrysemys picta) were sampled from wetlands exposed to chronically high microcystin levels due to a prolonged HAB event, and from unexposed, control wetlands. Physiological stress levels and several measures of immune functioning were compared between the HAB-exposed and control populations. Physiological stress levels, measured as heterophil:lymphocyte ratios, were higher in barn swallows, red-winged blackbirds, and Northern watersnakes exposed to a chronic HAB compared to unexposed, control individuals, but painted turtles did not differ in physiological stress levels between HAB-exposed and control individuals. Neither barn swallows nor red-winged blackbirds differed in immune functioning between populations, but HAB-exposed watersnakes had higher bactericidal capacity than control snakes, and HAB-exposed painted turtles had lower bactericidal capacity than control turtles. These results suggest that even when HABs do not cause direct mortality of exposed wildlife, they can potentially act as a physiological stressor across several taxa, and furthermore may compromise immune functioning in some species.

Hypoxia-inducible factor-1α (HIF-1α) plays a critical role in immune and inflammatory responses and is important in controlling a variety of processes in monocytes and macrophages. However, the role of HIF-1α in the teleost immune system remains less known. In this study, we cloned the cDNA sequence of HIF-1α from the ayu (Plecoglossus altivelis, PaHIF-1α). Sequence and phylogenetic tree analysis showed that PaHIF-1α clustered within the fish HIF-1α tree and was closely related to that of Northern pike (Esox lucius). PaHIF-1α was expressed in all tested tissues and expression increased in liver, head kidney, and body kidney upon Vibrio anguillarum infection. PaHIF-1α was found to regulate the expression of cytokines in ayu monocytes/macrophages (MO/MФ). PaHIF-1α mediated hypoxia-induced enhancement of MO/MФ phagocytic and bactericidal activities to enhance host defenses. Compared with the control, intermittent hypoxia further increased the expression of PaHIF-1α mRNA, improved the survival rate, and reduced the bacterial load of V. anguillarum-infected ayu. Therefore, PaHIF-1α may play a predominant role in the modulation of ayu MO/MФ function.

In a number of taxa, males and females both display ornaments that may be associated with individual quality and could be reliable signals to potential mates or rivals. We examined the iridescent blue/green back and white breast of adult tree swallows (Tachycineta bicolor) to determine whether plumage reflectance is related to adult or offspring immune responses. We simultaneously addressed the influence of blood selenium levels and the interaction between blood selenium and plumage coloration on adult and nestling immunity. Selenium is a well-known antioxidant necessary for mounting a robust immune response but its importance in wild birds remains poorly understood. In females, the brightness of white breast coloration was positively associated with bactericidal capacity, but there was no association with blood selenium. In contrast, male bactericidal capacity was associated with an interactive effect between dorsal plumage coloration and blood selenium concentration. Males with bluer hues and greater blue chroma showed increased bactericidal capacity as blood selenium concentrations increased, while bactericidal capacity declined in greener males at higher blood selenium concentrations. In nestlings, bactericidal capacity was positively associated with nestling blood selenium concentrations and white brightness of both social parents. These results suggest that white plumage reflectance is indicative of quality in tree swallows and that greater attention should be paid to the reflectance of large white plumage patches. Additionally, the role of micronutrients, such as selenium, in mediating relationships between physiology and signals of quality, should be explored further.

Phytohemagglutinin (PHA)-induced swelling is widely used to investigate cell-mediated and innate immunity across different vertebrate taxa. However, its physiological mechanism is still an open question due to the complexity of the involved immune components. In the present study, we measured the synchronous variations of PHA response, the proportion of different subtypes of leukocytes, as well as serum bactericidal capacity in circulation blood at 6, 12 and 24 h after PHA versus PBS injection in striped hamster, Cricetulus barabensis. First, the results showed that PHA responses reached a peak at 6 h postinjection, then sharply declined at 12 h and 24 h postinjection. Serum bactericidal capacity was higher at 6 h and 12 h than at 24 h. The proportion of different subtypes of leukocytes, as well as the ratio of neutrophils to lymphocytes did not display significant changes across different time points. Second, PHA response was positively correlated with the proportion of neutrophils and serum bactericidal capacity. The proportion of monocytes was negatively correlated with that of eosinophils and neutrophils. The proportion of basophils was negatively correlated with that of lymphocytes. Our results indicate that earlier enhanced PHA response is important for the striped hamster to cope with changing environmental conditions due to its small body mass, and the increased components of innate immunity in circulation blood may contribute to the enhancement of PHA swelling response.

Anthropogenic activities often produce pollutants that can affect the physiology, growth and reproductive success of wildlife. Many metals and trace elements play important roles in physiological processes, and exposure to even moderately elevated concentrations of essential and non-essential elements could have subtle effects on physiology, particularly during development. We examined the effects of exposure to a number of elements from a coal fly ash spill that occurred in December 2008 and has since been remediated on the stress and immune responses of nestling tree swallows. We found that nestlings at the site of the spill had significantly greater blood concentrations of Cu, Hg, Se and Zn in 2011, but greater concentrations only of Se in 2012, in comparison to reference colonies. The concentrations of elements were below levels of significant toxicological concern in both years. In 2011, we found no relationship between exposure to elements associated with the spill and basal or stress-induced corticosterone concentrations in nestlings. In 2012, we found that Se exposure was not associated with cell-mediated immunity based on the response to phytohaemagglutinin injection. However, the bactericidal capacity of nestling plasma had a positive but weak association with blood Se concentrations, and this association was stronger at the spill site. Our results indicate that exposure to these low concentrations of elements had few effects on nestling endocrine and immune physiology. The long-term health consequences of low-level exposure to elements and of exposure to greater element concentrations in avian species require additional study.
